Lipid Metabolism: Ceramide Transfer Protein Adds a New Dimension

چکیده
Just as non-lipidologists were becoming reconciled to the complexities of networks of ceramide metabolism and the diverse roles of ceramide in cell regulation and membrane structure, a new study reveals a new layer of regulation of ceramide through the actions of a ceramide binding/transfer protein.

New Aspects of Silibinin Stereoisomers and their 3-O-galloyl Derivatives on Cytotoxicity and Ceramide Metabolism in Hep G2 hepatocarcinoma Cell Line
Ceramide as a second messenger is a key regulator in apoptosis and cytotoxicity. Ceramide-metabolizing enzymes are ideal target in cancer chemo-preventive studies. Neutral sphingomyelinase (NSMase), acid ceramidase (ACDase) and glucosyl ceramide synthase (GCS) are the main enzymes in ceramide metabolism. Silymarin flavonolignans are potent apoptosis inducers and silibinin is the most active com...

متن کاملRegulation of secretory transport by protein kinase D–mediated phosphorylation of the ceramide transfer protein
Protein kinase D (PKD) has been identified as a crucial regulator of secretory transport at the trans-Golgi network (TGN). Recruitment and activation of PKD at the TGN is mediated by the lipid diacylglycerol, a pool of which is generated by sphingomyelin synthase from ceramide and phosphatidylcholine. The nonvesicular transfer of ceramide from the endoplasmic reticulum to the Golgi complex is m...
متن کاملAlternation in the Glycolipid Transfer Protein Expression Causes Changes in the Cellular Lipidome
The glycolipid transfer protein (GLTP) catalyzes the binding and transport of glycolipids, but not phospholipids or neutral lipids. With its all-alpha helical fold, it is the founding member for a new superfamily, however its biological role still remains unclear. We have analyzed changes in the HeLa cell lipidome in response to down- and up-regulation of GLTP expression. We used metabolic labe...
متن کاملA study on the targeting of ceramide metabolism by (-)-epicatechin gallate, catechin and quercetin in A-549 lung cancer cell line
Catechin, epicatechin gallate (ECG) and quercetin, as bioactive flavonoids, have been shown to possess anticarcinogenic effects. Ceramide plays an important role in killing tumor cells. Accordingly, the aim of this study was to clarify the involvement of these compounds in ceramide metabolism in A549 cancerous cell line. Spectrophotometer, cell culture and HPLC methods were used. Cell viability...
